The in vitro mammalian cell gene mutation test is used to detect mutations of the hypoxanthine-guanine phosphoribosyltransferase (HPRT) gene in Chinese hamster ovary (CHO) or lung (V79) fibroblasts. 

  • Cultures are incubated with several doses of the test compound for 4 hours in the presence and absence of S9 metabolic activation system

  • Cells are subcultured for 8 days after treatment to allow expression of the mutant phenotype, and then plated in media with and without 6-thioguanine (TG) to select for mutants and determine cloning efficiency.

  • The mutant frequency is determined by seeding known numbers of cells in a medium containing the selective agent in order to detect mutant cells and in a medium without the selective agent in order to determine the cloning efficiency. After an appropriate incubation time, colonies are counted.

  • A positive outcome is characterized by a statistically significant, dose-dependent increase in mutant frequency that exceeds historical negative control limits.
