HEPSPAN™ LONG-TERM CULTURE PRIMARY HEPATOCYTE MEDIUM AND HEPSPAN™ LONG-TERM CULTURE PRIMARY HEPATOCYTE
HEPSPAN™ LONG-TERM CULTURE PRIMARY HEPATOCYTE MEDIUM
Current standard monolayer hepatocyte cultures are typically functional in culture for 5 to 7 days. There is an increasing need for longer term cultures of hepatocytes to study liver toxicity, drug metabolism, drug interactions, viral infection, inflammation, liver development and regeneration. In order to extend the functional life of hepatocytes in culture, we have developed HepSpan™ medium, a long-term culture of primary hepatocyte medium which maintains the morphology and function of primary hepatocyte in standard 2D cultures for 4 weeks (human hepatocytes), 3 weeks (rat hepatocytes) and 2 weeks (mouse hepatocytes).
HepSpan™ medium is optimized for primary hepatocyte to enhance cell viability, function and long-term maintenance. HepSpan™ is a better representation of the liver’s extracellular environment for the initiation and maintenance of primary hepatocytes long-term cultures.
HepSpan™ Medium Advantages
Better representation of the liver’s extracellular environment
Improved hepatocytes function and high levels of liver-specific gene expression
Stable albumin and urea secretion for four weeks in culture
Maintain Long-term CYP activities and CYP inducibility of primary hepatocytes
Support long-term culture of cryopreserved and fresh plateable primary hepatocytes
HepSpan™ Medium Features
Defined medium tailored to support hepatocyte long term in vitro culture
Adjusted to standard 2D monolayer culture and high throughput
Raw material screening and quality control ensure minimal lot-to-lot variability
Multi-species culture medium available (Human, Rat, Mouse, Dog, Monkey)
HepSpan™ Medium Application
Hepatic Disease Model
Drug Metabolism
Metabolic Disorders Study

HepSpan™ Ready-to-Use Long-Term Cultured Primary Hepatocyte
Primary hepatocytes and their adherent cultures are the gold-standard in vitro tool to evaluate liver toxicity, drug metabolism, cytochrome P450 induction, drug-drug interactions, viral infection, inflammation, liver development and regeneration. However, primary hepatocytes rapidly lose their morphology and liver-specific functions, such as detoxification, activity of phase I and phase II enzymes, and the production of plasma proteins like albumin over a few days in standard culture.
MB Biosciences also provide HepSpan™ Ready-to-use long term cultured primary hepatocytes which include a pre-seeded hepatocytes in multi-well formats plates with long-term culture medium to overcome the short stability culture problem as well as standard issues of working with primary hepatocyte such as hepatocyte plating failure, poor viability, and alleviates any unnecessary time and cost spent on finding exceptional lots.
HepSpan™ ready-to-use cultured primary hepatocytes with a prolonged culture up to 4 weeks for human hepatocytes, 3 weeks for rat hepatocytes and 2 weeks for mouse hepatocytes in standard 2D-culture formats (24 wells, 48 wells, 96 wells and 384 wells).
HepSpan™ ready-to-Use Hepatocytes is an innovative platform for ADME/TOX, viral infection, inflammation and liver biology study. They are stored and transported at room temperature, and enables the ability to perform the experiments that require a longer time culture of primary hepatocytes.
